Not a peptide — a methylquinolinium small molecule. 5-Amino-1MQ blocks nicotinamide N-methyltransferase (NNMT), the enzyme that consumes methyl groups and nicotinamide to make 1-methylnicotinamide. Shut that reaction down and the cell stops bleeding methyl donors into a metabolic dead end.
The research interest follows directly from the chemistry: inhibit NNMT and labs ask what happens to the NAD+ salvage pool and to SAM-dependent methylation downstream. That is the question most of the published work is built around.
The compound is studied mostly as a probe of NNMT itself: how potently it occupies the enzyme, and what that occupancy does to the NAD+ salvage pool, SAM-dependent methylation, and adipocyte metabolism. Mitochondrial and cardiac NAD+ work has been added more recently.
As an NNMT inhibitor, 5-Amino-1MQ is used in work that treats the methylation–NAD+ junction as a metabolic lever. Cell and tissue models examine what happens when the enzyme is taken out of the loop — chiefly the effect on the NAD+ salvage pathway and on adipocyte energy handling.
The structure–activity paper from Neelakantan and colleagues anchors the chemistry; later studies extend it into NAD+ homeostasis and mitochondrial readouts, building the case for NNMT as a tractable metabolic target.
Interest in the molecule starts with the enzyme. Once NNMT was tied to obesity-related metabolic dysfunction, the field went looking for cell-permeable inhibitors, and methylquinolinium scaffolds — 5-Amino-1MQ among them — came out of that medicinal-chemistry effort.
Work since has used it largely as a tool compound: a clean way to ask what a cell does when NNMT activity drops.
